Recent studies have demonstrated that various nonsteroidal anti-inflammatory drugs (NSAIDs) induce hyperalgesia in various tissues. There is growing evidence that protein kinase C (PKC) plays an important role in nociception. Our aim was to determine whether indomethacin induces hyperalgesia and whether PKC is involved in its mechanism of action. We measured the antinociceptive effect of indomethacin in the second phase of the formalin test in rats. Hyperalgesia was induced by intrathecal (i.t.) injection of H-89 (a specific inhibitor of protein kinase A). We showed that i.t. injection of indomethacin (0.4-40 microg) produced dose-dependent hyperalgesia and that pre-treatment with indomethacin (i.t. or i.v.) decreased the antinociceptive effect of i.t. morphine. H-89 (10 nmol) induced hyperalgesia and blocked the antinociceptive effect of indomethacin (i.t.). Our results show that indomethacin induces hyperalgesia by activating the PKC pathway.Integrated combustion systems are widely used for producing gases that can be used in a variety of ways.
